What's the easiest way to create a Lower Third in AE6.5 for export to Premiere without having to render?

If this is this a motion Lower 3rd, there is no such thing as "no rendering". You can Export a quicktime, but it is still rendering a movie.

If you don't want to render, you bought the wrong program.

If you put aside the rendering, what is the best way to create a motion graphic lower third with alpha channel to be used in Premiere?

Any type you create in AE will have an alpha channel, you just need to render it out in a codec that supports an alpha, shuch as Animation. DV will not.
